The aim of the project is to unveil the heterogeneity of OA, with a special focus on the inflammatory subtype, by investigating its unique cellular composition, molecular pathways, epigenetic patterns, and interaction with microbiome composition. The ultimate goal is to identify specific diagnostic markers and therapeutic targets for this particular OA subtype.

This will be achieved through the following specific objectives:

Objective 1: To elucidate the OA heterogeneity and identify a molecular, cellular, and epigenetic profile of the iOA subtype, comparing tissues obtained from knee replacement surgeries between iOA and noninflammatory OA patients.

Objective 2: To identify a subset of iOA-related molecular, metabolic, and epigenetic markers in the synovial fluid and blood samples.

Objective 3: To establish a link between the immunological hallmarks of iOA and the gut microbiome, complemented by an analysis of systemic and local microbial, molecular, and metabolic markers.
