Characteristics and genetic diversity studies of pathogenic and relatively pathogenic mycobacteria present in Latvia

Latvian Council of Science research grant #09.1286.

Project manager: Viesturs Baumanis

The project provides further genotyping of M.tuberculosis and other Mycobacterium isolates, as well as to analyze the molecular mechanisms of drug resistance-causing with particular attention to second-and third-line drugs.

  • genotyping by restriction fragment polymorphism analysis - so called spoligotyping; to fully establish for use short tandem repeat analysis, which is considered as reference method since 2010;
  • to approbate the possibility to genotype isolates of Mycobacterium  from archaeological material;
  • localize mutations that cause resistance to kanamycin, p-aminosalicylic acid and fluoroquinolones;
  • complete the development of quick detection method of  fluoroquinolone resistance and approbate this on unknown clinical material;
  • in collaboration with the National Lung Diseases and Tuberculosis agency integrated in National Center of Infectology, as well as Latvian University of Agriculture to gather information on the collected and described isolates, and make recommendations for antiepidemiologic actions and providing a rational therapy.

The study will result in faster detection of drug resistance. Genotyping would provide possibility to document the transfer of infection and to take more effective preventive activities.
